Does anyone know if it will securely adhere to plexiglass?

I'd think so but who knows. Maybe try a test spot? Then you can just remove it if it doesn't work out. Or possibly you could add something for it to adhere to like primer or cement glue. You could try asking /diy/ first if no one else here knows.

Why not just use caulk though?

I want to build a 4-sided structure about 5' tall in a pyramid shape...i will adhere tiki masks to the sides and the fill in the gaps will be spray foam painted to resemble a volcano. All this is going to serve as a fountain so it must be waterproof.
